Thiol-containing amino acids homocysteine (Hcy) and cysteine (Cys) play important roles in regulated various biochemical pathways, whereas the tripeptide glutathione (GSH) is a major determinant of redox status. The ability to quantify each of these thiols is critical for understanding their role in pathophysiological processes (1) . This is a very difficult task because of the relatively low intercellular concentrations of free homocysteine, cysteine and the ease with which they undergo redox cycling with cellular glutathione disulfide and protein disulfides. We have developed a method for the quantification of these thiols using prior derivatization with the specific reagent 4-fluro-7-sulfamoylbenzofurazan (ABD-F) (2). Subsequent quantification using stable isotope dilution methodology makes it possible to analyze these molecules with high precision and accuracy.
