Depletion effect of High-abundance Urine Proteins for Detecting Markers of Bladder Cancer in Urine by Quantitative Mass Spectrometry

Bladder cancer is the most common malignancy in urinary tract. An accurate biomarker not only helps physicians to estimate the risk of recurrence but also reduce the number of cystoscopy performed which is invasive and costly. Several marker candidates of bladder tumor discovered from urine and cells have been reported. However, a urine-based assay does not hold superior properties in both sensitivity and specificity currently. Concentration dynamic range of urinary proteins exists and suppresses the detection of minor proteins. No commercial product for depletion of high-abundance urine proteins is available currently. We developed a facile method for depleting the three most abundant urinary proteins. The condition was optimized to study the different urine proteomes of bladder cancer by quantitative MS.
