Liquid chromatography-triple quadruple mass spectrometry (LC-MS/MS), a superior analytical technique for quantifying small molecule drugs, has been applied to quantitation of proteins in complex biological matrixes. Its main advantage over ligand binding assays (LBAs) is capability to detect and quantify unique peptides in specific regions of the proteins for overcoming interference from anti-antibody response. In addition, it provides wider linear range and improves assay precision. To develop a generic approach for quantitation of monoclonal antibodies (mAb) in human serum, two unique surrogate tryptic peptides of mAb were detected in multiple reaction-monitoring (MRM) mode. The result shows that LC-MS/MS assay enabled us to achieve lower limit of quantitation of 195 ng/mL and better accuracy within ±10% for the two unique peptides.
