摘要

A new 96 well plate based high throughput mass spectrometric method for the analysis of free NNAL, NNAL-N-Gluc and NNAL-O-Gluc in small urine volumes from smokers has been developed. The accuracy and precision of the method was determined and the assay was applied to more than 2600 samples for each analyte from a study of smokers from five ethnic groups. In studies where urine volumes are not limited we have used 250 μL of urine with reagents scaled accordingly to produce results with better assay LODs. The basic method should be applicable to studies of smokers, smokeless tobacco users and second hand smoke to further define the utility of these metabolites as potential biomarkers of cancer susceptibility.
