LODs for the direct analysis of oligonucleotides have been determined. HPLC-ESI MS can achieve lower LODs than MALDI-TOF MS for the direct analysis of oligonucleotides. For the analysis of siRNA sized oligonucleotides: (1) Using MALDI-TOF MS gave an LOD of between 0.15-0.68 fmol. (2) Using HPLC-ESI MS an LOD of 0.03 fmol was obtained.
