Hydrogels are widely used for controlled release applications because of their soft tissue biocompatibility, the ease with which the drugs are dispersed in the matrix and the high degree of control achieved by selecting the physical and chemical properties of the polymer network. Hydrogels consist of polymer chains crosslinked to each other to create a tangled mesh structure, providing a matrix for the entrapment of drugs. Chitosan, a cationic polysaccharide, is obtained by alkaline deacetylation of chitin, the principal exoskeletal component in crustaceans1. Chitosan is reported to be non-toxic and bioabsorbable2, and has been explored for the release of many drugs because of its mucoadhesivity and ability to enhance the penetration of drugs3-4. The swelling kinetics and release properties of crosslinked chitosan and chitosan/polymer blending matrices have been reported, the effect of molecular parameters of chitosan on controlled release property has also been studies, but the reported studies on the effect of chitosan acid salt on controlled release property was few. So the aim of this study was to prepare three chitosan hydrogels based on three typical kinds of acid, widely used as solvent of chitosan, to observe their morphology and structure by SEM, and to investigate the influence of acid salts on swelling behavior and controlled release property of chitosan hydrogels in simulating periodontal pocket environment using tetracycline HCl widely used in periodontitis treatment as model drugs.
