Purpose: For screening IOP lowering effects of new drugs several animal models are in use but most involve invasive techniques and are expensive. We evaluated the pharmacological utility of the rat model of steroid-induced ocular hypertension (SIOH) by studying the time course of IOP lowering by single drop application of 5 classes of antiglaucoma drugs. Methods: SIOH model was developed by bilateral topical application of dexamethasone 0.1% twice daily for 40 days after baseline IOP estimation. A rise of 20% from baseline was considered significant and these animals were included in the study. IOP lowering effect of single drop application of pilocarpine 2%, timolol 0.5%, dorzolamide 2%, brimonidine 0.2% and latanoprost 0.005% was evaluated in five groups of SIOH rats (n=8). In each group, 10mul of test drug was instilled in one eye and collateral eye served as control. IOP was estimated at regular intervals over 24 hours post-instillation. Results: Significant IOP elevation (pO.OOl) was observed in 80% of the dexamethasone treated animals at day 40. Single drop application of pilocarpine caused mean peak IOP reduction of 16.25% at 1.5 hours post-instillation. Timolol, dorzolamide and brimonidine produced mean peak IOP reduction of 19.66, 19.20 and 17.30%, respectively at 2 hours post-instillation. Peak effect of latanoprost was observed at 10 hours post-instillation and amounted to 26.91%. Conclusion: The study demonstrated that the rat model of SIOH is susceptible to changes in aqueous humor dynamics involving several different mechanisms and, therefore, is of significant pharmacological utility in screening investigational antiglaucoma agents.
