Schizandrin B isolated from the fruit of Schisandra chinensis, has been reported as an anti-cancer substance. In this study, we investigated the effects of Schizandrin B on the tumor suppressor gene NF1 in AMLl-ETO-expressing cell lines. We found thatAMLl-ETO-expressing cell lines displayed a low level of NF1 gene. Chromatin immunoprecipitation assays demonstrated that NF1 was the direct transcriptional target of AML1-ETO. The universal binding of AML1-ETO to genomic DNA resulted in recruitment of methyl-CpG binding protein 2 (MeCP2), indicating that AML1-ETO induced heterochromatic silencing of NFL Our findings indicate that Schizandrin B can relieve NF1 suppression in AMLl-ETO-expressing cells through a mechanism that involves inversion of epigenetic alterations.
