Low Temperature Requiring Strains of Paddy Straw Mushroom, Volvariella volvacea and Markers for Their Identification

The present study emphasized on developing low temperature requiring strains for fruiting in Volvariella volvacea. A total of 11 strains were evaluated for their fruiting at 22-26℃ using chemically treated paddy straw and composted cotton ginning mill waste, followed by studying its relationship with variabilities in ITS sequences, thermal death points and the activities of mycolytic enzymes (chitinase, N-acetyl-β-glucosaminidase and protease). On chemically treated paddy straw, only strains WV-3 of Volvopluteus earlei, Vv-11 and Vv-12 of V. volvacea fruited at bed temperature range of 23-26℃, with the highest yield by WV-3. In bigger scale trials (40 kg compost/bed) at 22-26℃, strain Gw-01 fruited the earliest (11. 93 days post-spawning) and produced the highest yield of 30. 79 kg/100 kg dry substrate on composted cotton ginning mill waste, followed by strains Vv-13 and Vv12. The remaining eight strains gave very low yields. Strain GVv-01 giving the highest yield at low temperature conditions is distinct from low yielding strains in the dendrogram deduced from the ITS sequences. Together with data from a previous study using four strains, the fruiting ability at low temperature was not found to have any correlation with activities of chitinase and N-acetyl-βglucosaminidase recorded on storage of cultures at refrigerated conditions. However, the activity of protease was recorded to be the lowest in strain Vv-11 giving the second highest pinning after strain GVv-01, while highest in strain Vv-09 that produced no fruiting at low temperature conditions. Thermal death point of low temperature requiring strains Gvv-01 and Wv-03 was found to be the lowest, followed by Vv-13 and Vv-11 under dry conditions and it may have some relationship with the temperature requirement of fruiting in these strains. The study has implications in finding low temperature requiring strains for fruiting and the development of markers for their identification.
