The effects of load on the secretion and expression ofrnbone markers were tested at different stages ofrndifferentiation of primary human osteoblasts.rnNHOs were both seeded with and without cytodex 3rnbeads (Sigma),transferred to a NASA rotating wallrnvessel (modeled microgravity) and harvested at day 7rnand day 14. Differentiated and undifferentiated NHOsrnwere loaded at 6-50G for 30 min and compared to cellsrnincubated at 1G after 1 day and 3 days. Collectively thernresults demonstrate that load has a differential effect onrnosteoblast differentiation as seen in modeledrnmicrogravity and shows specificity in expression ofrnbone cell markers vs. expression of secreted paracrinernsignaling markers.
