虎杖提取物对CCl4致建鲤肝损伤抗氧化系统的影响

         

摘要

主要探讨基础饲料中添加虎杖提取物对建鲤肝损伤中抗氧化性能的影响.建鲤被随机分为6个组,每组20条,即Ⅰ组(CCl4)、Ⅱ组(0.5%虎杖提取物)、Ⅲ组(1%虎杖提取物),按每10 g体质量0.05 mL体内注射30% CCl4;Ⅳ组(0.5%虎杖提取物)、Ⅴ组(1%虎杖提取物)体内不注射CCl4,只注射同体积植物油;Ⅵ为空白对照组,也注射同体积植物油.每日饲喂2次,连续8周,注射72 h后模型建立成功.检测建鲤血清中LDH、GPT、GOT、TP、ALB的表达水平;检测肝组织匀浆中SOD、GSH-Px、MDA、CAT、T-AOC含量的变化.结果表明:1%的虎杖提取物(Ⅴ组)对于清除自由基,防治肝损伤有很好的效果,能显著提高SOD、GSH-Px含量,降低MDA含量;显著提高T-AOC和CAT含量;显著增加TP、ALB活性;显著降低血清中GPT、GOT、LDH的活性,与空白对照组相比,差异不显著(P>0.05);与CCl4组相比,差异显著(P<0.05),而0.5%和1%虎杖提取物对照组则无明显变化.%This study focused on the effcts of oxidation resistance of basal diet added with polygonum cus-pidatum extract against Jian carp liver injury. Jian carps were randomly divided into six groups , I(CC14) , II (0.5% polygonum cuspidatum extract) , III (1% polygonum cuspidatum extract) group were injected (ip) with 30% (v/v) of CCl4 in arachis oil at a volume of 0. 005 mL/g of body weight; IV(0. 5% polygonum cuspidatum extract) and V (1% polygonum cuspidatum extract) control group had no CCl4 injection, according to the weight injected the same volume of arachis oil; VI was the blank control group, according to the weight only injected with the same volume of arachis oil. Twice daily feeding was conducted for 8 weeks. After 72 h, the expression levels of LDH, GPT, GOT, TP, ALB in Jian carp serum were detected; and the activities of SOD, GSH-Px, of MDA, CAT, and T-AOC in liver homogenates were also detected. The results showed that: 1% polygonum cuspidatum extract had a good effect on clearing the free radicals and treating the liver injury. It could significantly improve the content of SOD, GSH-Px, decrease the content of MDA, significantly improve the content of T-AOC and CAT, significantly increase the activity of TP, ALB, significantly increase the activity of GPT, GOT, LDH. Compared with the blank control group, no significant difference (P>0.05); compared with the CC14 group, differences were significant (P <0. 05) was observed. But the 0. 5% and 1% Polygonum cuspidatum extract control groups showed no significant change.
