益生乳杆菌的筛选研究

         

摘要

以分离保存的15株乳杆菌为研究对象,从中筛选益生乳杆菌两株.通过检测乳杆菌的疏水性和自聚性,得到表面疏水性和自聚性均较高的菌株为Ind-3、CH10和M8.其中,乳杆菌CH10和M8在模拟胃肠环境下活菌数均达到106 mL-1以上.通过灌胃乳杆菌CH10和M8,表明小鼠体重增加明显高与对照组,且实验组小鼠粪便中的β-半乳糖苷酶活性明显高于对照组.灌胃两株乳杆菌后,小鼠肠道内双歧杆菌与乳杆菌活菌数提高了,且肠杆菌、肠球菌和产气英膜梭菌的生长繁殖受到不同程度的抑制,而对照组无明显变化.结果表明筛选出的两株乳杆菌可以促进小鼠对营养物质的吸收,提高肠道内有益菌的数量,抑制有害菌的生长繁殖,或许能够改善寄主肠道的微生态环境.%15 Lactobadlli strain swere used for the screening of probiotic bacteria, in which 2 strains, i.e., Lactobadllus saiivarius CH10 and Lacto-bacillus murinus M8, showed potential probiotic properties. In vitro the hydrophobicity and auto-aggregation of these candidate bacteria were tested to find three strains out of 15 lactobacilli had high surface hydrophobicity and auto-aggregation, i.e., strain Ind-3, strain CH10 and strain M8. Simulation of gastrointestinal conditions revealed that only the viable cells of strains CH10 and M8 exceeded over 106 mL-1 at pH 2.5 and in the presence of 0.3% bile salt after incubation at 37℃ for 6 h. Mice fed with strain CH10 and M8 got higher body weight than the control. The mice fed with strains CH10 and M8 had higher β-galactosidase activity in their fecal, and the live numbers of lactobacilli and bifidobacteria in their intestinal gut were promoted. However, no significant changes in β—galactosidase activity, and the live numbers of lactobacilli and bifidobacteria in the control were observed. Meanwhile, the growth inhibition of Entaobaclena, Entaococcus and Clostridium perfnngens from the intestinal micro—flora occurred after the mice were orally administrated with strains CH10 and M8 for 10 days. Both Lactobacillus strains CH10 and M8 screened in this study would modulate the ecological environment of the intestines in mice due to the improvement of beneficial bacteria and inhibition of the harmful bacteria. They should be promising as probiotic candidates for further utilization due to the promotion of the body weight in mice model.
