Objective To establish a solid phase extraction ( SPE )/ LC-MS/MS method for determination of resveratrol in blood. Methods Made selfmade extraorion cartridges, added about 0. 02g glass sponge in the bottom of a 13 centimeters long acid burette,then followed by the bottom to the top added about 0. 15g of PSA,0. 3g of Florisil and 0. 45g of Silica column packings. Resveralrol were extracted from human blood using selfmade extraction cartridges and detected by LC-MS/MS. Qualitative and quantitative analysis was obtained by retention time (Rt) and Multiple Reaction Monitoring ( MRM) of LC-MS/MS. Results The recovery was 95. 66% -98. 58% . Limits of Detection (LOD) was 1. 79μg/mL. The linear range was 2. 5 ~ 20μg/mL. The correlation coefficient was 0. 9997. Conclusion The method is applicable to the analysis of resveratrol in blood.%目的 建立血液中白藜芦醇的固相萃取/LC-MS/MS方法.方法 自制固相萃取柱,在长约13cm的酸性滴定管中,底部填人约0.02g玻璃棉,依次加入柱填料PSA 0.15g,Florisil 0.3g,Silica 0.45g.对样本进行固相萃取,采用LC-MS/MS方法进行检测,运用保留时间和多反应监测(MRM)方式对血液中白藜芦醇进行定性定量分析.结果 回收率为95.66%~98.58%,最低检测限(LOD)为1.79μg/mL,线性范围2.5~ 20μg/mL;相关系数为0.9997.结论 本文所建方法,适用于血液中白藜芦醇的检测.
