白细胞介素23在哮喘小鼠动物模型中表达的研究

         

摘要

Objective:To study the expression of IL-23 in model of asthma in asthmatic mice. Methods:Mouse models were established and divided into two groups, normal control group and asthma group. The levels of IL-23 in serum were detected by ELISA, the levels of IL-23pl9mRNA and IL-23pl9 protein in lung tissue were detected respectively by RT-PCR and immunohistochemistry. Spleen T lymphocytes were isolated and cultivated in vitro. The levels of IL-23pl9mRNA and IL-23pl9 protein in T lymphocytes were detected respectively by RT-PCR and Western blot. Results:Compare to normal control group, the level of IL-23 in serum was higher in asthma group (P<0. 01) , the levels of IL-23pl9mRNA and IL-23pl9 protein in lung tissues were higher in asthma group(P<0. 01) , and the levels of IL-23pl9mRNA and IL-23pl9 protein in spleen T lymphocytes was higher in asthma group(P<0. 01) . Conclusion: IL-23 may involve in the pathogenesis of asthma, and may have influence on T lymphocytes.%目的:研究IL-23在哮喘小鼠模型中的表达.方法:构建动物模型,分为正常组、哮喘组,应用ELISA法检测两组小鼠外周血IL-23水平、RT-PCR法检测肺组织中IL-23p19mRNA表达、免疫组化法检测肺组织中IL-23表达;分离小鼠脾脏T淋巴细胞进行体外培养,应用RT-PCR法检测小鼠脾脏T淋巴细胞内IL-23p19mRNA表达水平、Western blot检测IL-23p19蛋白表达情况.结果:血清IL-23水平较正常对照组高(P<0.01),肺组织中IL-23p19mRNA及IL-23蛋白表达均较正常组高(P<0.01),哮喘组脾脏T淋巴细胞内IL-23p19mRNA及IL-23p19蛋白表达水平均较正常组高(P<0.01),有统计学意义.结论:哮喘小鼠中,IL-23在血清、肺组织、脾脏T淋巴细胞中表达均较正常小鼠明显增高,提示IL-23参与哮喘发病,且可能通过调控T淋巴细胞的功能而发挥作用.
