Objective:To establish the method for mass concentration of lobaplatin in human plasma and hydrothorax by HPLC.The method was used for patients with lung cancer,and could provide basis for the clinical therapeutic drug monitoring(TDM).METHODS: Human plasma and hydrothorax samples were centrifuged and extracted with acetonitrile.The Inertsil-ODS-SP column(250 mm×4.6 mm,5 μm)was used,the mobile phase were consisted of phosphate buffer(pH=6.5)and acetonitrile(containing 0.03%triethylamine)(V∶V=96∶4)and flushed%目的:建立同时测定人血浆、胸水中洛铂质量浓度的高效液相色谱法,并应用于肺癌患者,为临床治疗药物监测提供依据.方法:人血浆、胸水样品离心后,经乙腈蛋白沉淀,色谱柱为Inertsil-ODS-SP色谱柱(250 mm×4.6 mm,5 μm),流动相为pH=6.5的磷酸盐缓冲液-乙腈(含0.03%三乙胺)(V∶V=96∶4),流速为1.0 ml/min,于215 nm波长处进行检测.结果:胸水中洛铂质量浓度在0.1~80.0 μg/ml、血浆中洛铂质量浓度在0.1~50.0 μg/ml范围内呈现良好线性关系,洛铂在胸水、血浆中最低定量限和检测限分别为0.1、0.5 μg/ml.结论:本研究建立了快速测定人胸水、血浆中洛铂浓度的高效液相色谱法,该方法灵敏、稳定、重现性好、选择性好、准确度高,可用于准确测定人胸水、血浆中洛铂的含量,并用于治疗药物监测,有利于对患者进行个体化治疗.
