气相色谱法测定蝉花中角鲨烯的含量

         

摘要

Objective] To establish a method of capillary gas chromatography to detect the content of squalene in Cordyceps cicadae.[Method] Squalene in Cordyceps cicadae Shing was detected by gas chromatography.And the squalene contents in different parts of C.cicadae were com-pared.The optimal condition for the detection of squalene in C.cicadae was obtained.[Result] C.cicadae (1 g) was extracted by ultrasonic ex-traction,the product was methyl esterified at 40 ℃for 5 min by 1 mL of 0.5 mol/L sodium hydroxide-methanol solution and then extracted by n-hexane.The squalene could be determined quantitatively by GC.When the squalene concentration was from 21.43 to 214.30 μg/mL,there was a good linear relation (r=0.999 2) and a perfect method precision (RSD =3.71%),and the recovery rate was 77.80%-97.32%,and the aver-age recovery rate was 88.10% (n=9).[Conclusion] This method was simple and rapid with low pollution and high sensitivity,and it is suitable for determination of squalene content in C.cicadae.%[目的]建立毛细管气相色谱( GC)测定蝉花中角鲨烯含量的方法。[方法]采用气相色谱法测定蝉花中角鲨烯,并对不同部位天然蝉花的角鲨烯含量进行比较,确定蝉花中角鲨烯含量测定的最佳条件。[结果]1 g蝉花经超声波提取后,经1 mL 0.5 mol/L的氢氧化钠-甲醇溶液40℃皂化5 min,再经正已烷萃取净化,采用GC法测定角鲨烯含量。该方法在角鲨烯质量浓度为21.43~214.30μg/mL (r=0.9992)范围内线性关系良好,方法精密度良好(RSD =3.71%),加标回收率为77.80%~97.32%,平均加标回收率(n=9)为88.10%。[结论]气相色谱法简单快速,污染小,检测灵敏度高,可用于蝉花中角鲨烯含量的测定。
