大黄素对实验性自身免疫性心肌炎大鼠IFN-γ及IL-17水平的影响

         

摘要

目的 探讨大黄素对自身免疫性心肌炎大鼠IFN-γ及IL-17的影响,及其对大鼠的心肌保护作用.方法 6周龄雄性Lewis大鼠24只随机分为3组(8只/组):自身免疫性心肌炎(EAM)组、大黄素(Emodin)组和对照(Control)组.于实验0d及7d,EAM组和Emodin组用心肌肌球蛋白/完全弗氏佐剂免疫,而对照组仅用完全弗氏佐剂假性免疫.Emodin组于0d起,每日给予50 mg大黄素灌胃治疗,共21 d,而EAM组和对照组每日给予蒸馏水灌胃.21d时处死大鼠,心肌组织行HE染色及Masson染色,光镜下评价心肌组织炎细胞浸润及纤维化程度;Western blot法检测心肌组织IFN-γ及IL-17蛋白的表达水平;Elisa法检测血清中IFN-γ及IL-17的水平.结果 与EAM组相比,大黄素显著减轻了Emodin组心肌组织中的炎性细胞浸润及纤维化改变.EAM组和Emodin组血清及心肌组织中IFN-γ和IL-17的水平明显高于Control组(P<0.05).而Emodin组上述指标均明显低于EAM组(P<0.05).结论 大黄素可以降低IFN-γ和IL-17的水平,对EAM大鼠的心肌起到保护作用.%Objective To investigate the effects of emodin on the IFN-γ,IL-17 expression in EAM rats and its protective effect on myocardial.Methods Male Lewis rats (n =24,6 weeks old) were randomLy assigned into three groups (8 animals/group):experimental autoimmune myocarditis (EAM) group,emodin (Emodin) group and control (Control) group.On days 0 and 7,EAM group and Emodin group were immunized with myosin/Freund's complete adjuvant (FCA),the Control group was immunized with FCA alone.Emodin group was intragastric administrated with emodin 50 mg/kg per day for 3 weeks,while EAM group and Control group were given distilled water.All rats were sacrificed on day 21.Myocardial tissue were stained with hematoxylin and eosin (HE) and Masson's,and analyzed under a light microscopy to evaluated the degree of inflammatory cell infitration and fibrosis;Westem blot analysis assay was performed to detect IFN-γ and IL-17 protein expression levels in myocardial tissue;Serum concentration of IFN-γ and IL-17 was measured with ELISA kits.Results Emodin significantly reduced inflammatory cells infiltration and fibrosis in Emodin group compared to EAM group.IFN-γ and IL-17 levels in serum and myocardial tissue were markedly increased in EAM group and Emodin group compared to the Control group (P < 0.05).Moreover,the above mentioned indexes in Emodin group were obviously lower than EAM group (P < 0.05).Conclusion Emodin can reduce the levels of IFN-γ and IL-17,and protect the myocardial tissue in EAM rats.
