Objective To investigate the effect of Rapamycin ointment(RAP)on atopic dermatitis in mice and its possible mechanisms. Methods DNCB was repeatedly applied on the back of NC/Nga mice to establish the animal model of atopic dermatitis. 50 mice were divided in-to 3 groups:control group,model group and RAP group(0.05%RAP,0.1%RAP and 0.2%RAP),and 10 mice each group. RAP lasted for 14 days. Levels of several cytokines,including interleukin(IL)-4 and IL-5,interferon(IFN)-γ,Immunoglobulin E(IgE)from the serum samples were detected by Elisa kit at 8 days and 15 days. Meanwhile,Skin samples underwent the measurement of thickness and weightness at 8 days and 15 days as well. Results RAP could significantly decrease the skin thickness and weightness at 8 days and 15 days. Meanwhile,RAP markedly inhibited IL-4,IL-5,IgE,and increased the level of IFN-γ from the serum samples at 8 days and 15 days. Conclusion The ther-apeutic effect of RAP on atopic dermatitis-like skin lesions may be taken through inhibiting IgE,IL-4 and IL-5 synthesis and promoting the se-cretion of IFN-γ.%目的 探讨雷帕霉素软膏(RAP)对Nc/Nga小鼠特应性皮炎的保护作用及其相关机制的研究.方法 利用2,4-二硝基氯苯(DNCB)建立Nc/Nga小鼠实验性特应性皮炎模型.50只小鼠分为5组:正常对照组、模型组、实验组(又分为0.05% RAP组、0.1% RAP组、0.2% RAP组),每组10只.实验组连续用药3周,分别于第8天、15天测量各组小鼠背部皮损厚度和皮肤质量,并利用ELISA法分别检测第8天、15天血清中细胞因子干扰素γ(IFN-γ),白介素4(IL-4),白介素5(IL-5),免疫球蛋白E(IgE)的表达.结果 雷帕霉素软膏治疗第8天、15天,皮损厚度及皮肤质量明显下调(P <0.05),且各实验组血清中细胞因子IL-4、IL-5和IgE水平明显低于模型组,IFN-γ水平明显高于模型组(P <0.05),且呈剂量依赖性.结论 雷帕霉素软膏可能通过促进IFN-γ分泌,下调炎症因子IL-4、IL-5水平以及抑制IgE表达,从而对小鼠特应性皮炎产生保护作用.
