绵羊miR-1基因前体序列的多态性分析

         

摘要

To study the single nucleotide polymorphism in pre-miR-1 among sheep breeds with different meat production characteristics and analyze its association with skeletal muscle development, an asymmetric PCR-SSCP technique was used to detect SNP of pre-miR-1 sequences in sheep breeds with different meat production characteristics, including Suffolk sheep and Hu sheep, and the effect of SNP sites on the secondary structure and stability of pre-miR-1 was predicted.The experimental results showed that an A→G mutation was detected at 115 bp downstream of the mature sequence of miR-1, and the SNP locus had a certain correlation with the meat production performance of sheep.In Suffolk sheep, an excellent mutton sheep breed imported from abroad, GG is the dominant genotype.The G allele frequency reaches 0.76.The prediction of the secondary structure of pre-miR-1 indicated that the polymorphism site caused a change in the secondary structure of pre-miR-1 and theΔG decreased by 1.5 kJ/mol.In conclusion, the SNP in pre-miR-1 may have a significant impact on the processing of miR-1, and affects the performance of sheep meat production.%为了检测不同产肉性能的绵羊群体中miR-1基因前体序列的单核苷酸多态性(Single nucleotide polymorphism,SNP),并与绵羊骨骼肌发育进行相关性分析,采用不对称聚合酶链式反应-单链构象多态性(PCR-SSCP)技术对萨福克羊和湖羊2种产肉性能不同绵羊的miR-1基因前体序列进行SNP检测,并预测该SNP位点对miR-1前体序列二级结构稳定性所造成的影响.结果表明,在绵羊miR-1基因前体成熟序列的下游115 bp处检测到1个A→G突变,并且该SNP位点与绵羊的产肉性能有一定的相关性;由国外引进的优良肉羊品种萨福克羊,GG基因型为其优势基因型,G等位基因频率为0.76;通过miR-1基因前体序列二级结构预测发现,该SNP位点能够使其发生变化,使ΔG降低1.5 kJ/mol.综上所述,绵羊miR-1基因前体序列的SNP位点可能对miR-1前体的加工成熟产生重要影响,最终影响绵羊的产肉性能.
