Using potato plantlets in vitro Zhengshu 5 as materials, effects of sucrose, polyethylene glycol ( PEG6000) and mannitol on growth and conservation of plantlets were studied. The results showed that the three osmotic regulators mentioned above at certain concentrations inhibited the growth of plantlets but did not lower the conservation rates. Of the osmotic regulators, 40 g · L-1 PEG6000 had the biggest inhabitation. RAPD analysis showed that the plantlets conserved on the medium supplymented 90 g · L-1 sucrose, 40 g · L-1 PEG6000 and 20 g · L-1 mannitol did not exhibit somaclonal variation.%以“郑薯5号”马铃薯试管苗为材料,研究蔗糖、聚乙二醇( PEG6000)和甘露醇对试管苗生长和保存的影响.结果表明,3种渗透调节物质均能抑制试管苗生长,但不影响试管苗的存活率,其中40 g·L-1 PEG6000的抑制效果最好.采用RAPD标记研究发现,保存于添加90 g·L-1蔗糖,40 g·L- PEG6000和20 g·L-1甘露醇的培养基上的试管苗没有发生无性系变异.
