屋尘螨提取液对哮喘气道上皮细胞间质转化的影响

         

摘要

目的 探究屋尘螨提取液(House dust mite extracets,HDM)对人支气管上皮16-HBE细胞间质转化(Epithelia-mesenchymal transition,EMT)的影响.方法 以16-HBE细胞株为研究对象,分别加入HDM和转化生长因子β1(Transforming growth factor-β1,TGF-β1),应用MTT法和Western blot法,检测细胞增殖情况以及EMT过程中E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)和α-平滑肌肌动蛋白(α-SMA)表达.结果 ①HDM刺激后,16-HBE细胞形态改变不显著,同时对细胞增殖无显著影响;与TGF-β1联合作用后,细胞形态发生EMT变化,并且显著促进细胞增殖(P<0.05).②HDM刺激对16-HBE细胞中E-cadherin、Vimentin和α-SMA蛋白表达均无显著影响;与TGF-β1联合作用后,E-cadherin蛋白表达显著降低(P<0.05),Vimentin和α-SMA蛋白表达升高(P<0.05),效果较TGF-β1单独应用更为明显.结论 HDM不能直接刺激上皮细胞发生EMT改变,但可通过促进TGF-β1诱导的上皮细胞EMT改变而发挥作用.%Objective To explore the role of house dust mite extracts (HDM) in the epithelial-mesenchymal transition (EMT) of human bronchial epithelial (16-HBE) cells.Methods Human bronchial epithelial cells (16-HBE) were treated with HDM and transforming growth factor-β1 (TGF-β1).After treatment for 72 h,the morphology and proliferation capacity of 16-HBE cells were investigated with light microscope and ~ method,and the expression of E-cadhedn,Vimentin and α-SMA was tested by western blot.Results ①Treatment with HDM did not induce significant morphological change and proliferation of 16-HBE cells;HDM combined with TGF-β1 induced significant morphological changes and proliferation of 16HBE cells (P < 0.05).②Treatment with HDM induced no significant change in E-cadherin,vimentin and α-SMA expression,but the expression of E-cadherin,vimentin and α-SMA changed significandy after the combination use of HDM and TGF-β1 (P < 0.05).Conclusion HDM has no significant effect on EMT process of 16-HBE,but it can significandy enhance TGF-β1-induced EMT process.
