金钱草胶囊血药浓度测定方法的建立与验证

         

摘要

Objective: To establish a rapid, sensitive and specific method for determining the plasma concentration of Lysimachia capsule. Methods: HPLC method was used, the column type was Kromasil C8, and different mobile phase compositions, propor-tions and concentrations were used to screen mobile phase. The optimal mobile phase, methanol-0.4% phosphorous acid (50:50) was selected. The flow rate was 1.0mL/min. The detective wave-length was 360nm and column temperature was 25oC. Results: The effective ingredient of Lysimachia is quercetin. Its retention time is 8.8min. There was a good linear relationship between the concentration and peak area integral value in the range of 0.8~16μg/mL for Plasma Concentration of Lysimachia cap-sule(r=0.9988). Precision test RSD=6.06% (n=6). The average recovery rate was 98.01% (RSD=1.21%) (n=6). Conclusion: The method is accurate, simple and good reproducibility, which can be used for Plasma Concentration of Lysimachia capsule and pharmacokinetic and bioavailability studies.%目的:建立一种快速、灵敏、专属的方法测定金钱草胶囊血药浓度。方法:采用高效液相色谱法,以Kromasil C8柱为分析柱,采用不同的流动相组成、比例、浓度等对流动相进行筛选。以甲醇-0.4%磷酸溶液(50:50)为流动相,流速1.0mL/min,检测波长为360 nm,柱温为25℃。结果:金钱草中有效成分槲皮素的保留时间为8.8 min左右,槲皮素血浆浓度在0.8~16μg/mL范围内峰面积与浓度有良好线性关系(r=0.9988),精密度试验RSD=6.06%(n=6),方法灵敏度高,LOD为0.08μg/mL,LOQ为0.8μg/mL。平均回收率98.01%(RSD=1.21%)(n=6)。结论:本方法测定结果准确、简便,重现性好,可用于金钱草胶囊血药浓度的测定和药动学、生物利用度等的研究。
