Thecircling(cir/cir)mouseisamurinemodelforhumannonsyndromicdeafnessDFNB6.Transmembraneinnerear(tmie)isthecausativegeneanditsmutationthroughdeletionofa40-kilobasegenomicregionincludingtmieleadstodeafness.ThefunctionofTmieisunknown.TobetterunderstandthefunctionofTmie,wefocusedonthespatiotemporalexpressionoftmieintheratcochleabyusingaTmie-specificantibody.Resultsshowedthattmieexpressionwasprominentinearlypostnatalratcochleasinthestereociliabundlesofhaircells.TheTmiesignalspreadfromthestereociliatothehaircellbodyregionandontoorganofCorticells.NoTmiesignalwasobservedincellnuclei;Tmiewaslocalizedtothecytoplasm.BecauseTmieispredictedtohave1or2transmembranedomains,wepostulatethatitislocalizedtomembrane-basedorganellesortheplasmamembrane.OurresultsimplythatTmieexistsinthecytoplasmandmayhaveakeyroleinthematurationandstructureofstereociliabundlesindevelopinghaircells.Afterhaircellmaturation,TmieisthoughttobeinvolvedinthemaintenanceoforganofCorticells.
