CD13isamembrane‐boundectopeptidase,highlyexpressedonmonocytes,macrophages,anddendriticcells.CD13isinvolvedindiversefunctions,includingdegradationofpeptidemediators,cellularadhesion,migration,viralendocytosis,signaling,andpositivemodulationofphagocytosismediatedbyFcγRsandotherphagocyticreceptors.Inthiswork,weexploredwhetherbesidesactingasanaccessoryreceptor,CD13byitselfisaprimaryphagocyticreceptor.WefoundthathCD13mediatesefficientphagocytosisoflargeparticles(erythrocytes)modifiedsoastointeractwiththecellonlythroughCD13inhumanmacrophagesandTHP‐1monocyticcells.TheextentofthisphagocytosisiscomparablewiththephagocytosismediatedthroughthecanonicalphagocyticreceptorFcγRI.Furthermore,wedemonstratedthathCD13expressioninthenonphagocyticcelllineHEK293issufficienttoenablethesecellstointernalizeparticlesboundthroughhCD13.CD13‐mediatedphagocytosisisindependentofotherphagocyticreceptors,asitoccursintheabsenceofFcγRs,CR3,andmostphagocyticreceptors.PhagocytosisthroughCD13isindependentofitsenzymaticactivitybutisdependentonactinrearrangementandactivationofPI3KandispartiallydependentonSykactivation.Moreover,thecross‐linkingofCD13withantibodiesrapidlyinducedpSykinhumanmacrophages.Finally,weobservedthatantibody‐mediatedcross‐linkingofhCD13,expressedinthemurinemacrophage‐likeJ774cellline,inducesproductionofROS.TheseresultsdemonstratethatCD13isafullycompetentphagocyticreceptorcapableofmediatinginternalizationoflargeparticles...
