Derivation of attenuated porcine epidemic diarrhea virus (PEDV) as vaccine candidate

The field isolate of porcine epidemic diarrhea virus (PEDV) was serially passaged in Vero cells. The cell passaged PEDV, designated KPEDV-9, was tested for its pathogenicity in neonatal pigs, immunogenicity and safety in the pregnant sows. 53 and 11piglets, respectively were i.m. inoculated with 106.0 and 107.0 TCID50/ml of cell adapted PEDV. None of thes piglets had diarrhoea or symptoms related to PEDV. Eight piglets were infected orally with 10 ml of virus stock which contained virus at 108.0 TCID50/ml and were artificially fed with dairy milk. Three pigs showed signs of anorexia and diarrhoea in two or three days, but these signs were transient. One of 3 pregnant sows was inoculated with the attenuated virus and showed increased immune responses by ELISA. In addition, one group of 10 and two groups of 5 delivered piglets were shown to be protected from challenge with wild type PEDV when compared with the challenge of one group of 10 and one group of 5 control piglets. The safety test involving 63 pregnant sows indicated that all inoculated animals farrowed the average numbers of litters of piglets. It is concluded that the attenuated virus derived from serial passage could be applied as a vaccine for suckling piglets against PEDV.
