In 1944, DNA was characterized as the carrier for genetic information. The ultra-structure of DNA and the mechanisms of gene duplication during cell division were subsequently uncovered in 1953 by Watson, Crick and others. Over decades' research on DNA then primarily focused on how genetic information was encoded, conserved and passed on. Unexpectedly, an additional, non-informative role of nucleic acids was discovered around the turn of the century: foreign nucleic acids derived from pathogens were identified as molecular patterns which could be sensed by the host in bacterial or viral infections. Pathogen-derived DNA or RNA is internalized by specialized host immune cells and detected within the endosomal compartments by membrane-bound toll-like receptors (TLRs 3, 7, 8 and 9). TLRs then initiate an innate immune response leading to the secretion of type I inter-ferons and proinflammatory cytokines.
