In the present study, differential expression of key DNA sequences responsive to salt stress in plant seedlings subjected to 150 mm NaCl, was investigated in a salt-sensitive canola line (Cyclone) by differential display reverse transcription PCR (DDRT-PCR). A canola cDNA fragment (SCA(1)H(27)) was isolated using an arbitrary primer that was up-regulated under stress, but showed no similarity to any of known plant genes from the nucleotide database through BLAST analysis. However, it showed significant similarity to the chloroplast DNA sequences thereby confirming its similarity. The predicted amino acid sequence of the isolated cDNA sequence showed significant similarity to the carcinoma-related and the catalase-peroxidase protein sequences of species other than Brassicaceae using the SwissPort and PDB protein database. The results showed the supposition of induction of carcinoma-related DNA sequences in the chloroplast of plant cells under salt stress. Further proteomic studies would be helpful in identifying definite function of this novel canola cDNA upregulated in relation to salt sensitivity.
