Purification and characterization of desmutagenic components extracted from sweet potato leaves

A major desmutagenic factor, purified from water extract of sweet potato (Ipomoea batatas Lam, CN1489-89) leaves by ultrafiltration and gel filtration, and identified using Salmonella typhimurium TA98, was found to have a molecular weight of 269,000and to be made up of protein (42.18 percent) and sugar (57.40 percent). It also exhibited an absorption peak at 280 nm with a small shoulder in the range of 320-330 nm, suggesting that it might be a glycoprotein. The antimutagenicity of the novel substance was studied against 2-amino-3-methylimidazo-[4,5-f]-quinoline (IQ) and its activated metabolites. The desmutagenicity of the novel substance against lQ and its activated metabolites were 58 and 63 percent, respectively and the induced mutation frequency reduced from 5,500 X 10~(-9) to 3,400 X 10~(-9) reflecting a 38 percent bio-antimutagenicity at the same concentration of 0.05 mg. It was found that the suppressive effect of the desmutagenic factor on mutation caused by IQ and its activated metabolites could be ascribed to both desmutagenicity and bio-antimutagenicity, although the former was higher than the latter.
