Stress-related hormonal and metabolic responses to restraint, with and without butorphanol administration, in pre-conditioned goats

Twelve goats were pre-conditioned to handling, restraint, instrumentation, clippers and the experimental room. On the day of the trial, goats were confined in a stanchion, with or without administration of butorphanol (0.1 mg/kg of body weight, i.v.), in a cross-over design experiment. Heart and respiratory rates were measured at baseline (0), 15, 30, 45, 60, 75, 90, 105 and 120 min. Rectal temperature was measured and blood samples were collected at baseline and 30, 60, 90 and 120 min. Epinephrine, norepinephrine, cortisol, glucose and free fatty acid concentrations were measured. At all times, glucose concentration was significantly lower when butorphanol, compared with saline, was administered. Baseline concentration of free fatty acids was significantly higher than that at all other times for either treatment. Differences for treatment or time were not significant for epinephrine, norepinephrine or cortisol values. There was a significant treatment and time interaction for respiratory rate, heart rate and rectal temperature. Respiratory rate was significantly higher at 60 min after butorphanol treatment, compared with saline treatment. Heart rate was significantly higher at 75 min than at baseline, and rectal temperature was significantly higher at 30 than 120 min. It is suggested that the analgesic efficacy of butorphanol can be more rationally evaluated in goats experiencing pain because the response in goats not experiencing pain is now known.
