Important discoveries that come as a complete surprise and cause radical intellectual reorientations in a field of investigation are not rare in modern biomedical research. In some ways, they actually represent a form of reward to those active in that field. The field of cellular responses to DNA damage (DDR) has had a fair share of such surprising discoveries. Thus, the Ku protein was considered an autoantigen with transcription factor properties before its actual function in the repair of DNA double strand breaks (DSBs) by non homologous end joining (NHEJ) was elucidated. The DNA dependent protein kinase catalytic subunit (DNA-PKcs) was an exotic protein kinase with unknown functions, showing a puzzling DNA requirement for activity, before its involvement in NHEJ and its interactions with Ku were characterized. And DNA Ligase IV was a biochemically unknown entity before the gene was cloned and its function in NHEJ elucidated.
