Glioblastoma multiforme are malignant primary tumors of the central nervous system that remain one of the most deadly cancers in adults. One recent concept proposes that cancer cells derive from a small fraction of self-sustaining cells with the exclusive ability to initiate and propagate the tumor. In brain tumors, glioblastoma stem-like cells (GSCs) might participate in resistance to radiotherapies and contribute to recurrence and aggressiveness. Conversely, reducing the GSC reservoir slows tumor progression in animal models.Cancer stem cells, including GSCs, are believed to lodge in a defined micro-environment. Indeed, almost 20 years since the term "stem cell niche" was coined, a large body of work has supported its importance in the regulation of stem cell renewal and fate. Notably, this niche feeds the malignant stem-like subpopulation through locally secreted factors and metabolites. Similarly to embryonic and adult stem cells, cancer stem cells segregate within a specific and confined niche that contains a vascular structure. In the case of glioblastoma, GSCs have been identified surrounded by endothelial cells. Moreover, GSCs have been reported to differentiate toward the endothelial lineage and compose tumor vasculature.
