Neospora caninum causes abortion in cattle. Neosporavaccines can minimize and control the infection in mice but not in cattle. Eight mice were subcutaneously injected 3070% water-in-Montanide ISA 773 oil emulsions with 1 x 10~6 inactivated tachyzoites. After immunization and mating, pregnant mice were challenged with live 2x10~6 tachyzoites. Parasites present in the brain and tissues of the pups and adult mice were detected/assessed by PCR, serological and footpad tests. Vaccinated adults or pups showed no symptoms of neosporosis until euthanasia whereasun-vaccinated oil controls (8) showed severe weight loss and posterior limb paralysis. Four of eight plugged mice in the vaccinated group gave birth with lower pregnancy rate compared with controls. Antibody against N. caninum was detected in all mice vaccinated with whole killed tachyzoite compared with mice in control group. Vaccinated mice showed significant footpad swelling whereas controls did not and parasite DNA was detected in brain tissue of all controls but none in vaccinated mice.
