In 2008, the National Toxicology Program (NTP) published findings that chronic (2-year) exposure to hexavalent chromium [Cr(VI)] in drinking water resulted in increased incidences of oral mucosa tumors in rats and intestinal tumors in mice. Although informative to potential hazards of Cr(VI) ingestion, the concentrations employed in this study create uncertainty as to the relevance of these findings to humans. Importantly, data in the NTP study suggested that intestinal tumors occurred at concentrations that induced tissue damage—suggesting involvement of a mode of action (MOA) based on chronic tissue wounding and healing. Despite an otherwise dearth of data on the effects of Cr(VI) in the intestine, some regulatory agencies concluded that Cr(VI) induced intestinal cancer via a mutagenic MOA; thus employed a conservative extrapolation procedures in setting safe exposure levels to Cr(VI) which will dramatically impact site cleanup approaches for Cr(VI). To investigate the MOAs underlying these tumors, and their relevance to humans, a series of studies were designed to inform the MOAs for tumors of the intestinal and oral mucosae.
