New method to examine plasmodium metabolism in situ in the mosquito developed. Distinct differences in the phospholipid and FA profile of infected vs control midguts. Infected midguts display an increase in pro-inflammatory FA content, particularly AA. Imaging MS identified 28 species in higher abundance in the infected midgut. A distinct PE(38:5) species found localised to infection site. Tandem MS/MS analysis showed a 4:1 ratio of EPA vs AA at Sn2 position. EPA is a PUFA that suppresses immune response via oxidative metabolism products. Ongoing investigations are exploring the temporal changes in lipid content to better understand their roles.
