The complete proteome and the surface proteome of OMPC have been determined by tandem mass spectrometry. A total of 540 proteins have been identified with a predicted false positive rate of 0.5percent. OMPC obtained from a serogroup B strain (B11) is more readily characterized when compared to a serogroup C database (FAM18). ?The 10 most abundant OMPC proteins account for 75percent of the total protein signal intensity. PorA and PorB are the most abundant proteins in OMPC and account for approximately 50percent of the total protein signal intensity. Por A and Por B are only poorly accessible on the surface. The most surface accessible proteins of OMPC are rmpM and NadA. 344 proteins were found to have reacted with N-acetylhomocysteine thiolactone. Chemically reacted proteins account for >95percent of the total protein signal intensity. 36 predicted outer membrane proteins account for 75percent of the total protein signal intensity. OMPC Proteins that are not membrane associated appear to be nonspecifically attached to the OMPC surface. No evidence has been found for significant amounts of proteins that are trapped inside the OMPC. Chemical reaction efficiency of PorB Lysine residues are not driven by surface accessibility and can be only empirically determined. Factors such as, internal salt bridges, shielding by the membrane and shielding by oligomerization and protein complex formation seem to play an important role.
