One of the most promising ways to modulate the immune response is with gene therapy of the immune system (IS), including the use of DNA vaccines, which can generate an immune response after cell transfection. However, antigen-presenting cells (APC), the key-linkers between the innate and the adaptive immunity, are very resistant to transfection. To enhance the transfection efficiency of mouse APC, derived from the bone marrow or spleen, we used TATp-modified liposomal carriers (TATp-L) for the intracellular delivery of plasmids encoding for EGFP (pEGFP) or Bovine Herpes Virus I glycoprotein D (pCI-gD). Liposomes were also tested as DNA vaccine carriers. TATp-lipoplexes allowed for significantly enhancement of both the uptake and transfection of APC in vitro, and they were shown to be efficient in eliciting T lymphocyte mediated cytotoxicity in vivo after vaccination.
