Purpose: To study the effect of bile salt-lecithin mixed micelles on gastrointestinal epithelia from cell-based assay by Caco-2 cell model; Method:The mixed micelles was prepared by rotary evaporation method. The effect on cell viability, proliferation, integrity of gastrointestinal epithelia and apoptosis were evaluated by Caco-2 model. Result:The toxicity increased with the increase of concentration of mixed micelles. The mixed micelles with more than 0.4 mMaffected the cell viability and proliferation significantly when the ratio of lecithin to SDC was lower than 2:1. However, the effect on tight junctions of the mixed micelles with the SDC concentration between 0.2 mM and 0.6 mM were not damage but transient opening. The result of apoptosis also illustrated the mixed micelles with more lecithin induced less apoptosis; ConcIusions:The mixed micelles with the higher concentrations had certain toxicity to Caco-2 cell, while the toxicity declined with the increase of phospholipids.
