Objective To investigate the influence of using different kinds of tortoise shell on the content of bovine-hide gelatin from glue of tortoise shell. Methods The UPLC-MS method was performed on the Zorbax C18 column (50 mm × 2. 1 mm,1. 8 μm),by a gradient elution using 0. 1% formic acid aqueous solution-0. 1% formic acid in acetonitrile as the mobile phase at a flow rate of 0. 3 mL/min and at 40 ℃. Glue of tortoise shell were detected which were made from different kinds of tortoise shell. Results It was founded that the test results were positive which contained Trachemys scripta eleganns shell,while negative results were detected which were made by pure Chinemys reevesiis shell. Conclusion The method is simple,accucate,specific and high sensitivity which provided a basis for quality′s effective control of glue of tortoise shell.%目的：考察不同品种的龟甲原料对龟甲胶中牛皮源成分含量的影响。方法采用超高效液相色谱－质谱联用法（UPLC－MS），色谱柱为Zorbax C18柱（50 mm ×2.1 mm，1.8μm），以0.1%甲酸水－0.1%甲酸乙腈为流动相梯度洗脱，流速为0.3 mL/min，柱温为40℃。分别以不同品种龟甲为原料熬制龟甲胶，并对其牛皮源的含量进行测定。结果纯草龟龟甲熬制的龟甲胶检测结果为牛皮源阴性，掺杂巴西龟龟甲的龟甲胶检测结果为牛皮源阳性。结论该方法简便，专属性强，灵敏度高，可有效控制龟甲胶质量。
