To establish a HPLC method for determination of gentamicin in the raw milk. Protein in milk were mostly eliminated by the precipitators, OPA was used for derivatization, and then separated on C18 column with a mixed solution of methanol-acetic acid as mobile phase , and excitation and emission wavelength are 340 nm and 450 nm with a fluorescence detector The recovery were 85.6%-97.3%,the relative standard deviations (RSD) were 1.21%-4.04%, the minimum detectable values were 0.2μg/mL. This method can be used for the routine analysis of gentamicin in raw milk.%建立测定原料奶中庆大霉素C组分的高效液相色谱法.通过加入适量沉淀剂除去样品中大部分蛋白质,采用邻苯二甲醛试剂柱前衍生化,利用C18色谱柱,以含有0.02 mol/L的甲醇-醋酸溶液为流动相进行梯度洗脱,用荧光检测器检测,激发波长340 nm、发射波长450 nm.结果表明,回收率在85.6%~97.3%,精密度(RSD)为1.21%~4.04%,最低检出限为0.2μg/mL.该方法可用于原料奶中庆大霉素残留4种组分的测定.
