蛹虫草多糖抑制刀豆蛋白诱导的T细胞增殖

         

摘要

Objective To study the cell mechanism of Cordyceps Militaris extract inhibits immune reaction.Methods Mice splenocytes were seeded into U-shaped 96-well cell cuhure plate,the cell concentration was 3 × 105/well.0.8 μg/mL concanavalin A (ConA)was added into cell culture system and cultured for 3 days to stimulate T cell to proliferate,then added different concentration Cordyceps Militaris polysaccharides(CMP),and the concentration was 0.5 mg/mL and 1.0 mg/mL,respectively.Flow cytometry was used to detect the proliferation of CD4+ T cell and CD8+ T cell.Results CMP can inhibit the CD4+ T cell and CD8+ T cell proliferation induced by ConA.There were statistical significantly differences between CMP group and control group(P< 0.001),and it was with dose-effect relationship.Conclusion CMP can inhibit the T cell proliferation induced by antigen,and it is possible the cellular mechanism for CMP carrying out immune regulation function.%目的 探究蛹虫草多糖抑制免疫反应的细胞机制.方法 小鼠脾脏单细胞悬液接种于U型底的96孔细胞培养板,细胞接种密度为3×105/孔,采用0.8 μg/mL刀豆蛋白(concanavalin A,ConA)刺激3d,构建T细胞增殖模型.在培养体系中加入不同浓度的蛹虫草多糖(0.5 mg/mL和1 mg/mL),流式细胞学检测蛹虫草多糖对CD4+T细胞和CD8+T细胞的效应.结果 蛹虫草多糖可以抑制ConA诱导的CD4+T和CD8+T细胞增殖反应,与对照组相比,CD4+T细胞和CD8+T细胞增殖比例组间差异均具有统计学意义(P<0.001),且呈现剂量—效应关系.结论 蛹虫草多糖可以抑制抗原诱导的T细胞增殖反应,这可能是蛹虫草多糖发挥免疫调节效应的细胞学基础.
