中药多糖、胸腺肽对U14宫颈癌荷瘤鼠外周血TLC亚群及NK细胞的影响

         

摘要

目的 比较四种中药多糖及胸腺肽对U14宫颈癌荷瘤鼠肿瘤生长、外周血TLC、NK细胞的影响,试图找到对宫颈癌有相对特异性的免疫增强剂.方法 70只昆明小鼠,建立U14宫颈癌荷瘤鼠模型60只,设立正常对照组、肿瘤对照组、LBP、APS、GLP、PGE、胸腺肽组,每组10只,正常对照组、肿瘤对照组用生理盐水灌胃21 d、其他组分别用四种中药多糖和胸腺肽灌胃21d.第22天,所有动物处死后检测各治疗组的抑瘤率,流式细胞术比较各治疗组与正常和肿瘤对照组小鼠外周血CD4+CD8+TLC和CD49b+NK细胞亚群的改变.结果 肿瘤抑制率排序:胸腺肽<LBP<GLP,PGE和APS组与肿瘤对照组比较差异无统计学意义(P>0.05).肿瘤对照组外周血CD4+/CD8+TLC和CD49b+NK细胞与正常组比较均有所增高,但差异无统计学意义(P>0.05).LBP、GLP、胸腺肽组CD4+/CD8+TLC较正常组和肿瘤对照组明显升高,且差异有统计学意义(P<0.05),APS、PGE组CD4+/CD8+ TLC明显低于肿瘤对照组.各组外周血CD449b+NK明显升高,差异有统计学意义(P<0.05).结论 LBP、GLP和胸腺肽对U14宫颈荷瘤鼠有明显的免疫增强作用,但PGE、APS是否能用于宫颈癌的治疗,还需进一步研究.%Objective To compare the effect of four herbal polysaccharides and thymopeptide on tumor growth, TLC and NK cells in the peripheral blood of rats with U14 tumor, and try to find relatively specific immune in-tensifiers to cervical cancer. Methods Seventy Kunming rats were enrolled in this study, 10 of which were enrolled as the normal control group. The other 60 rats were constructed into rat models of cervical cancer, which were equally divided into 6 groups, namely tumor control group, LBP group, APS group, GLP group, PGE group and Thymopeptide group, each with 10 rats. The rats in the normal control group and tumor control group were given saline solution by gavage for 21 days, and the remaining five groups were given LBP, APS, GLP, PGE and Thymopeptide for 21 days, respectively. On the 22th day, all the rats were killed and tested for the tumor inhibitory rate. The CD4+CD8+ TLC and CD496+ NK changes in the peripheral blood were analyzed by flow cytometry and compared between the groups. Results The tumor inhibitory rate were increased gradually from the Thymopeptide group to the LBP group to the GLP group, but showed no statistically significant difference in the PGE group, APS group and tumor control group. The CD4+/ CD8+ TLC and CD496+ NK were increased in tumor control group than the normal control group, with no statistically significant difference. The CD4+/CD8+ TLC in the LBP group, GLP group and Thymopeptide group were significantly increased than that in the normal control group and tumor control group. The CD4+/CD8+ TLC in the APS group and PGE group were significantly lower than that in the tumor control group. The CD496+ NK in all the seven groups was significantly increased after treatment. Conclusion LBP, GLP and Thymopeptide exerts obvious immunity enhancement on U14 rats, but whether PGE or APS can be used in the treatment of cervical cancer should be further studied.
