纳豆激酶发酵液的脱色工艺研究

         

摘要

For decobrzation of nattokinase ferm entation liquor , resin 330 (OH ) was sejected from 11 different decolorzing medium s . The results showed that (1 )Restin 330 ( OH )could effectivey adsorb the cololring mattrrs in nattokinase ferm entation liquor. The kinetic model accorded to difflsing equation well . The kinetic polynom ial equalion was-ln (1 - F )=0 .0223 t+0 .0511 , R2 =0 .9978 , in which t was decolonizing time . (2 )Under static condition , the proportion of resin and ferm entation liquor was 15 g : 40 m L at pH 7 .0 for 200 min . The decolonzation effrciency w as 90% , the activiy of enzyme was 97 .67 and the purificaton m ultple was 3 .09 .(3 )The dynamic oondition consisted of pH 7.0 , flow-velocity of 1.0 BV/h and operation time 5h. The decolorzing liquor was mixed with eluant of phosphate buffer at pH 7.0 by flow-velocity 0.9 Bv. The total decoloization efficiency was 90% , the activity of enzyme was 98% and the purificaton multiple was 3 .41 .%从11种待选脱色介质中筛选出330(OH)型树脂对纳豆激酶发酵液进行脱色研究.结果表明:(1)330(OH)型树脂对纳豆激酶发酵液中的色素为优吸型吸附,其动力学模型符合扩散方程,其动力学拟和方程为-ln(1-F)=0.0223t+0.0511,R2=0.9978,其中F=Qt/Qe,Qt为脱色时间为t时330(OH)型树脂对色素的吸附量,Qe为330(OH)型树脂对色素的平衡吸附量.(2)在静态条件下,当pH 7.0,树脂与发酵液用量比为15 g(树脂):40 mL(发酵液)时,脱色200 min,其脱色率为90%,酶活保留率为97.67%,纯化倍数3.09;(3)利用该树脂进行动态脱色实验,当pH 7.0,流速为1.0 BV/h时,脱色5 h,用pH 7.0的磷酸缓冲液淋洗0.9 BV,将脱色液和淋洗液合并,其脱色率为90%,酶活保留率为98%,纯化倍数为3.41.
