KIF20 A基因在肝癌细胞及组织中的表达及临床意义

         

摘要

Objective:To investigate the expression of KIF20A in hepatoma cell lines,hepatocellular carcinoma tissues and paracancerous tissue ,and to analyze its clinical significance. Methods:RT-PCR and western blot were used to detect the expressions of KIF20 A in one normal liver cell line LO2 and seven HCC cell lines including QGY7701 ,HCC9810 ,QGY7703 ,HepG2 ,SMMC7721 ,Huh7 , Hep3B. Using the same methods,KIF20A expression in 5 pairs of HCC tissues and matched paracan-cerous tissues were examined. The Cancer Genome Atlas( TCGA)database was used to analyze the relationship of KIF20A expression with tumor grade and prognosis. Results:Protein and mRNA ex-pression of KIF20A were elevated in hepatocellular carcinoma cells and tissues. KIF20A expression significantly correlated with tumor grade and poor prognosis analyzed by TCGA. Conclusion:High ex-pression of KIF20 A is found HCC cell lines and tissues which were closely correlated with tumor grade and poor prognosis.%目的:探讨KIF20A在肝癌细胞系、肝癌组织及癌旁组织中的表达,并分析其临床意义。方法:采用RT-PCR和 western blot 检测正常永生化肝细胞 LO2和肝癌细胞 QGY7701、HCC9810、QGY7703、HepG2、SMMC7721、Huh7及Hep3B细胞内KIF2OA mRNA和蛋白的表达水平,同时检测KIF20A mRNA和蛋白在5对肝癌组织及相应癌旁组织的表达情况;通过癌症基因组图谱( TCGA)分析KIF20A表达与肝癌患者肿瘤分级和生存预后的关系。结果:KIF20A mRNA和蛋白在肝癌细胞中表达水平高于正常永生化肝细胞LO2中的表达,在肝癌组织中的表达水平高于配对癌旁组织( P<0.05);通过TCGA分析发现,KIF20A mRNA的表达与肝癌肿瘤分级及生存预后有关。结论:KIF20A在人肝癌细胞和肝癌组织中高表达,其高表达与肝癌肿瘤分级和不良预后有关。
