氟尿嘧啶明胶微球的体内释药特性的实验研究

         

摘要

Objective :To study the release characteristics of 5-Fu gelatin microsphere(FGM). Methods: FGM was perfused into the external carotid artery of rabbits by selective artery embolization under X-ray monitor, 5-Fu solution was infused into the external carotid artery and femoral vein as the controls. Blood and local tissue samples were tested at different time and analyzed statistically for drug concentration determination. Results: Higher blood and local drug concentration was produced immediatly after artery perfusion,kept for 20 min. and then decreased to 0 in 80 min. Heigh drug concentration kept from 40 to 90 min after slow injection into vein, there was no significant difference between circulating and local drug concentration. After artery embolization with FGM the local drug concentration was 20 times heigher than the blood drug concentration and the heigh level of local drug concentration kept for more than 140 min. Conclusion: External carotid artery embolization with FGM may significantly reduce the circulating drug level and increase the local drug concentration ,therefore ,may be used for the purpose of targeted tumor therapy.%目的:对氟尿嘧啶不同给药方法的效果加以对比,以了解氟尿嘧啶明胶微球在体内的释药特点。方法:在X线监控下,将氟尿嘧啶明胶微球超选择栓塞在兔颈外动脉及其分支,以氟尿嘧啶溶液灌注颈外动脉或在股静脉静滴作为对照,不同时间点取血样测试血药浓度,进行统计分析。取栓塞组织石蜡切片,常规HE染色,光镜观察。结果:动脉灌注法可以产生局部较高的血药浓度,但维持时间较短;静脉滴注法血药浓度维持时间较长,但局部与全身血药浓度无显著差异;使用氟尿嘧啶明胶微球进行栓塞可以在较小的用药剂量上在局部维持相对较长时间的较高的血药浓度。结论:氟尿嘧啶明胶微球进行动脉栓塞可以显著降低全身血药浓度及用药剂量,达到肿瘤靶向治疗的目的。
