二甲基精氨酸二甲胺水解酶1rs3087894C/G单核苷酸多态性与我国肝细胞癌遗传易感性的相关性

         

摘要

Objective To explore the relationship between dimelhylarginine dimethylaminohydrolase 1 ( DDAH1 ) rs3087894C/G polymorphisms and genetic susceptibility of hepatocellular carcinoma in Chinese population. Methods The genotyping of 120 cases with hepatocellular carcinoma( disease group) and 70 healthy persons( control group) was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method. Results The genotypic frequency of CC + CG,GG of DDAH1 rs3087894C/G were 33. 3% ,66.7% in disease group and 7. 1% ,92.9% in control group,respectively, there was significant difference of the genotypic frequency between the two groups( P <0. 05 ). Compared with GG genotype, the people who carried CC + CG genotye might have high risk to develop hepatocellular carcinoma ( OR = 3. 793 ,95% CI = 1.589-8. 307). The polymorphism of DDAH1 rs3087894C/G was related to hepatitis b virus infection and the level of alpha-fetoprotein(P<0. 05) ,but no correlation wilh sex.age, tumor size, the degree of tumor differentiation and lymph node metastasis(P>0.05). Conclusion The polymorphisms of DDAH1 rs3087894C/G is associated with the pathogenesy of hep-atoeellular carcinoma in Chinese population.%目的 探讨二甲基精氨酸二甲胺水解酶1( DDAH1) rs3087894C/G单核苷酸多态性与我国汉族人群中肝细胞癌(HCC)遗传易感性的关系.方法 用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)法对120例HCC患者(病例组)和70例健康对照者(对照组)进行基因型分析.结果 病例组DDAH1 rs3087894C/G位点CC+ CG、GG基因型频率分别为33.3%和66.7%,对照组基因型频率分别为7.1%和92.9%,2组比较差异均有统计学意义(P<0.05).与GG基因型相比,CC+CG基因型个体患HCC的风险度高(OR=3.793,95%CI=1.589 -8.307).DDAH1rs3087894C/G单核苷酸多态性与乙型肝炎病毒感染、甲胎蛋白水平有相关性(P<0.05),与患者性别、年龄、肿瘤大小、分化程度、淋巴结转移无相关性(P>0.05).结论 DDAH1 rs3087894C/G单核苷酸多态性与我国HCC发生有相关性.
