薄盖灵芝对反复热性惊厥大鼠海马IL-1和caspase-3表达的影响

         

摘要

目的 探讨薄盖灵芝对反复热性惊厥(FS)大鼠海马白细胞介素-1β(IL-1β)、白细胞介素-1受体拮抗剂(IL-1ra)与caspase-3表达的影响.方法 21日龄雄性SD大鼠随机均分为热性惊厥组(FS组),盐水对照组(NS组)及薄盖灵芝干预组(G组).采用热水浴诱导大鼠FS,隔日诱导1次,共诱导10次.G组于每次水浴前2h腹腔注射薄盖灵芝;而NS组腹腔注射相同体积的0.9%氯化钠溶液.末次热水浴后12h处死动物,用ELISA方法测海马IL-1β和IL-1ra的含量;RT-PCR法测海马IL-1β、IL-1ra及caspase-3 mRNA的表达水平.结果 G组大鼠海马IL-1β水平和IL-1βmRNA水平均明显低于NS组和FS组;G组大鼠海马IL-1ra水平及IL-1ra mRNA水平亦明显低于FS组和NS组;各组大鼠海马IL-1ra/IL-1β蛋白比值差别无统计学意义.G组大鼠海马caspase-3 mRNA水平均明显低于NS组和FS组.结论 薄盖灵芝能降低反复热性惊厥大鼠海马IL-1β和caspase-3表达水平,通过调节免疫及抑制凋亡发挥保护反复FS大鼠海马神经元的作用,而IL-1ra与该作用无关.%Objective To evaluate the effect of ganoderma capense on IL-1β and caspase-3 expressions in the hippocampus of rats with frequent febrile seizures(FS).Method 21-day-old male Sprague-Dawley(SD)rats were randomly divided into febrile seizure(FS)group, sodium chloride solution control(NS)group and ganoderma capense intervention(G)group. Febrile seizure was induced by hyperthermal bath,two hours before febrile seizures;rats in G group were given peritoneal injection of ganoderma capense,whereas the same volume of 0.9 % sodium chloride solution was injected into peritoneum of rats in NS group. IL-1β and IL-1ra levels in hippocampus of these rats were determined by ELISA methods;IL-1β,IL-1ra and caspase-3 mRNA levels in hippocampus were measured by RT-PCR.ResultsIL-1β mRNA and protein levels in hippocampus of rats in FS group were significantly lower than those in FS group and NS group. IL-1ra mRNA and protein levels in hippocampus of rats in FS group were significantly lower than those in FS group and NS group. IL-1ra/IL-1β protein ratios were no significant difference among groups. Caspase-3 mRNA levels in hippocampus of rats in FS group were significantly lower than those in FS group and NS group.Conclusions The neuroprotective effects of ganoderma capense may be associated with the downregulation of Caspase-3 and IL - 1β in the hippocampus of frequent FSs rats,but not IL-1ra.
