Thaumatin-like proteins (TLPs) from the seeds of sorghum (Sorghum bicolor) genotypes, namely APK 1, BSR 1, CO 26, CO 28, COS 28, TNS 30, TNS 342 and TNS 586, were extracted using cold extraction buffer. The protein extracts were concentrated by ammonium sulfate precipitation and then dialysed. The protein fractions showed significant activity against Alternaria sp., Rhizoctonia solani and Pyricularia grisea [Magnaporthe grisea]. The 50-80% ammonium sulfate precipitate from TNS 30 was further purified by DEAE-Cellulose column chromatography. The presence of TLP in the crude extract and the purified fractions was identified by western blot analysis using polyclonal anti-zeamatin antiserum. The molecular weight of this protein was found to be 22 kDa..
