Using an integrated computational and experimental systems biology approach, researchers have identified HIPK2 as a novel therapeutic target for kidney fibrosis. "We developed new tools that enabled us to identify upstream protein kinases responsible for altered gene expression in kidney disease," explains John He from the Mount Sinai School of Medicine.The researchers compared differentially expressed genes in the Tg26 HIV mouse model-which has both tubulointerstitial fibrosis and glomerulosclerosis-and wild-type littermates. By computationally identifying and ranking protein kinases potentially responsible for the observed changes in gene expression, HIPK2 was predicted to have an important role in HIV-associated nephropathy.
