Mature blades from Favorita, Xingjia No.2, Jingshanshu, GS393 and Zi 2 as the experimental materials, leakage ratio of electrolyte(REC), DAB reaction , alondialdehyde(MDA), soluble sugar contents under low temperature gradient processing were measured, coming to a comprehensive analysis method which closely related with phenotypic correlation of cold resistance, high repeatability, and convenient operation. The results showed that electrical conductivity method is easy to operate with simple technical requirements. The experimental results intuitively indicated that the frost resistance strength of each type. However, the disadvantage is that the electrolyte leakage rate of determination is influenced by environmental temperature, leading low data repeatability. Low instruments and personnel requirements are required to test Malondialdehyde (MDA) and soluble sugar contents, especially the well maneuverability. Its defect is that operation process is relatively complex, low repeatability and relatively complicated data processing compared with other methods. DAB reaction asks extremely low instrument, low researchers, simple operation and less artificial time. It can product results quickly and obvious phenotypic characteristics, simply identify the strength of the comparing the frost resistance of different varieties. At the same time, it is more convenient for frost resistance research with high repeatability and less interference factors.%将费乌瑞它、兴佳二号、金山薯、紫2作为试验材料,取成熟的顶端功能叶片进行低温梯度处理,分别进行电导率、过氧化氢DAB原位检测、丙二醛和可溶性糖含量分析,综合得出与耐寒表型相关性好、重复性高、方便操作的分析方法.结果表明:电导法简单便于操作,技术要求低,试验结果可以直观准确地表明各品种耐冻性强弱,缺点是电解质外渗率的测定受环境温度影响大,数据重复性较低;丙二醛及可溶性糖含量的测定所需仪器、人员等均要求低,可操作性强,其缺点是操作过程较繁杂,重复性较低,且数据分析过程相对较难;DAB组织定位法所需仪器、人员要求极低,操作简单,人工时间成本低,得到结果速度快,表型特征明显,易于识别与比较不同品种的耐冻性的大小,同时重复性强,干扰因素小,更方便用于耐冻性研究.
