槲皮素对乳腺癌MCF-7细胞电离辐射敏感性的影响

         

摘要

目的:探讨槲皮素对乳腺癌MCF-7细胞辐射敏感性的影响.方法:对体外培养的乳腺癌MCF-7细胞,随机分为A、B两组,A组42℃热休克处理2 h,诱导热休克蛋白表达,B组热处理前1 h加入终浓度为50、100、150 μmol/L的槲皮素,然后42℃热休克处理2 h.4 h后分别用X射线0、2、4、6、8、10、15 Gy辐照后继续培养.MTT比色法检测细胞增殖变化,克隆形成实验观察细胞存活率.取经A、B组(150 μmol/L)处理条件处理后的MCF-7细胞,采用流式细胞术Annexin V/PI双标法来测定细胞凋亡.结果:终浓度为50、100、150 μmol/L的槲皮素可以抑制MCF-7细胞增殖,对2~10 Gy照射剂量起到增敏作用.细胞克隆形成实验表明在实验浓度范围内,槲皮素可以降低细胞存活率,对X射线增敏作用有一定的浓度依赖关系,浓度越大,对X射线的增敏作用越强.流式细胞术Annexin V/PI双标法测定结果显示,槲皮素可以诱导MCF-7细胞凋亡增加.结论:槲皮素能够抑制乳腺癌MCF-7细胞增殖和克隆的形成,诱导细胞凋亡.%Objective: To investigate the influences on the sensitivity of Quercetin in ionizing radiation to breast cancer MCF-7 cells. Methods: MCF-7 cells were incubated in vitro and divided into two groups of A and B. Group A were incuxbaled at 42℃ for 2 h to induce expression of heat shock protein. Group B were pretreated for 1 h with 50, 100, 150 μmol/L Quercetin before heat treatment, and then treated with 42℃ for 2 h. Group A and B were irradiated with 0, 2, 4, 6, 8, 10,15 Cy X-ray after 4 h. Cell proliferation was detected by MTT method, cell survival rates were detected by clonogenic experimenc. The MCF-7 cells in group A and B (150 μmol/L) were taken to measure the cell apoptosis by Annexin V/PI double labled flow cytometry. Results: MCF-7 cell proliferation was inhibited and radio-sensitivity of 2 10 10 Gy X-ray was enhanced by 50, 100, 150 μmol/L Quercetin. Cell survival rates was reduced by Quercetin. The X-ray radio-sensitivity was depended on Quercetin concentration. The greater the concentration of X-ray increased, the susceptibility effect was stronger. Annexin V/PI double labled flow cytometry showed that, the percentage of MCF-7 cell apoptosis was increased by Quercetin. Conclusion: Quercetin can inhibit cell proliferation and colony formation, and can induce cell apoptosis of MCF-7 cells.
